Santa Cruz se joue en deux manches indépendantes où les joueurs colonisent une île en construisant des maisons, églises et phares, afin de récolter des ressources et donc d'accumuler des points. Le vainqueur est celui qui après la seconde Manche aura accumulé le plus de points.
